How to get casein?
About 80% of the total protein in cow's milk is casein. It is the main protein and is contained in the milk in the form of micelles and submicelles.
A micelle is a combination of casein molecules - submicelles, whose charged parts are directed outwards, and uncharged parts - inwards. The outer part of the micelle is hydrophilic, i.e. it is able to absorb water well, the inner one, on the contrary, is hydrophobic.
Being a protein of animal origin, casein has a complete amino acid composition. This means that it contains all 8 essential amino acids, of which all other conditionally replaceable and non-essential amino acids can then be synthesized.
Casein is obtained in 3 ways:
- rennet coagulation
- acid coagulation
- micro and ultrafiltration.
Rennet coagulation
Rennet is the process of turning casein micelles into a flaky mass. Merging, micelles form a dense gel (rennet casein), which also contains whey.
The casein obtained in this way is used to produce processed cheeses and cottage cheese. Rennet casein is insoluble in water.
Acidification
Acids and bases are used in this casein production method. In traditional casein precipitation (lactic acid precipitation), mesophilic microflora is used, i.e. non-gaseous bacteria.
When microflora is introduced into skimmed milk, it coagulates (clotting), i.e. loss of casein in the sediment, with its subsequent separation from the serum. With this method, get acid casein.
Caseinates are obtained by further processing of acid casein with a dilute alkali solution.
If sodium hydroxide solution is used in the process, sodium caseinate is obtained. When calcium hydroxide is used, alkalinization produces calcium caseinate.
Caseinates have not very good taste and rather poor solubility.
Filtration
For the production of high quality casein filtering method is used. In this production method, milk is not exposed to high temperatures and is treated with solvents and enzymes.
Micro- and ultrafiltration methods are gentle and preserve the natural structure of the protein.As a result of this method, micellar casein is obtained. Such casein has the best characteristics: solubility, taste, lack of allergy to it.
